The Purification of Plasmid DNA for Clinical Trials Using Membrane ...
ABSTRACTMembrane chromatography offers a good solution to the challenge of developing an efficient chromatographic procedure for plasmid DNA purification. The large convective pores of anion exchange membranes allow plasmid DNA to access all the anionic binding sites of the membrane at high flow rates. Here we demonstrate that the pIDKE2 plasmid can be purified from a recombinant Escherichia coli lysate using a Sartobind D membrane combined with size exclusion chromatography to render material with 95% purity and an average yield of 50%. This process yields therapeutically suitable plasmid [more...]

Plasmid DNA Recovery Using Size-Exclusion and Perfusion ...
Finally, a second size-exclusion chromatography step was carried out to purify the plasmid DNA from other small molecular-weight contaminants. Analytical methods proved that the purified plasmid DNA had a purity of 95% after Sephacryl S1000. Plasmid identity was confirmed by restriction enzyme digestion. Biological activity of the purified plasmid was confirmed in vivo; immunized mice developed a positive antibody response against all HCV structural antigens. This procedure offers an alternative to traditional methods that use organic reagents, mutagenic and toxic compounds, and [more...]

Quick Kit for Bacterial Total DNA Isolation
This kit is a very quick and easy one for bacterial total DNA (chromosome and plasmid) isolation with fewer steps. It takes less than 15 min. for sample to be ready for loading on agarose gel electrophoresis. Using this kit, the plasmid takes only one from its three forms after isolation, which a appears as one clear and sharp band in the agarose gel after electrophoresis (in addition to chromosomal band). Making the detection of plasmid in agarose is much easier and also determined, if the sample has more than one plasmid. This kit is able to isolate total DNA from bacteria cell with high [more...]

Variants of PCR (1)
The bacteriophage T4 DNA polymerase was also tested shortly after the first reports of PCR. It has a higher fidelity of replication than the Klenow fragment. Since it is also destroyed by heat, it has seen little use since the development of thermostable polymerases. The DNA polymerase from Thermus aquaticus (or Taq), was the first thermostable polymerase used in PCR, and is still the one most commonly used. The enzyme can be isolated from its 'native' bacterial source, or from a cloned gene expressed in E. coli. The Stoffel fragment is produced from a truncated gene for Taq polymerase, [more...]

Plasmid DNA Extraction Kit
DNA Extraction is the removal of deoxyribonucleic acid (DNA) from the cells or viruses in which it normally resides. Extraction of DNA is often an early step in many diagnostic processes used to detect bacteria and viruses in the environment as well as diagnosing disease and genetic disorders. The GenQuik Plasmid DNA Extraction Kit, utilizing an exclusive membrane technology in the form of convenient spin columns and 96 well plates, makes the isolation of high quality plasmid DNA from recombinant E.coli cultures simple, rapid and cost-effective. For Details on GenQuick Plasmid DNA [more...]
