Reveo's direct DNA sequencing using tunneling microscopy « The ...

Reveo is developing an ambitious technology to stretch out and deposit taut DNA on conductive surfaces for electronic base detection using one or more STM tips and tunneling current measurements. The linearization and deposition of nucleic acid sequences will likely be done using molecular combing. Reveo’s approach requires atomically flat and positively charged substrate surfaces (e.g. self-assembled monolayers on gold substrates or treated graphite substrates). In addition to molecular combing, Reveo has proposed other methods to linearize DNA, including electrophoretic and hydrodynamic [more...]

In Cancer, One Ring To Rule Them All

Imagine that a machine draws your blood, screens it for genetic mutations and chemical variations that can cause cancer, and then pops out a drug tailor-made for your DNA. The hypothetical drug would target and fix the point irregularities which have accumulated over time that can lead to the formation of tumors — and cancer. "Personalized cancer drug delivery? Depending on the approach, it could be as soon as 10 to 15 years away," says Viterbi professor Andrea Armani , an assistant professor of the Mork Family Department of Chemical Engineering and Materials Science. Armani has received [more...]

A GMO primer — What you should know about genetically modified ...

Ninety-five percent of the sugar-beet crop in the U.S. is genetically modified, but a recent federal ruling may have a huge impact on that. When it all began: The technologies that led to genetically modified foods were developed in the 1970s, with the advent of recombinant DNA technology. At the time, there were a lot of ethical debates and concerns about crossing the species barrier and transgenic technology. The focus then was on bacteria — mainly E. coli — and putting genes from different species into bacteria. Big Business: In the 1980s, Monsanto pushed to develop and use GM [more...]

Graphene could revolutionize DNA sequencing - physicsworld.com

By feeding individual strands of DNA through nanometre-sized holes, researchers in the Netherlands say they have proved the principle of a revolutionary new DNA sequencing technique. The breakthrough is part of a worldwide race to develop fast and low-cost strategies to analyse these codes that underpin the chemistry of life. The genetic profile – or "genome" – of an organism is determined by recording the full sequence of acid base pairs that make up its DNA. In 2003, the Human Genome Project made history by determining the entire human genetic code – 3 billion DNA base pairs that took [more...]

My Helical Tryst: Single Molecule DNA Sequencing

There are a number of companies that have recently launched competing technology platforms that are capable of sequencing DNA at the single molecule level. You may ask, why? Well, the answer is simply cost-effectiveness. DNA sequencing technology has been around for several decades, and to date, the Human Genome Project (HGP) is considered the pinnacle application of that technology. The cost of the HGP was approximately $3 billion and took about a decade and half. It was the molecular biology equivalent of putting a man on the moon. But what is the next step? Well, if we can systematically [more...]

Colony PCR: Another Use For Toothpicks « Promega Connections

Colony PCR: Another Use For Toothpicks July 26, 2010 by Isobel I still remember my JOY (sadly, yes) when I first learned about colony PCR. It allowed me to avoid a laborious procedure involving genomic DNA isolation, restriction digestion, and (the dreaded) Southern blotting. I was trying to show definitively that a transposon had successfully been inserted into a target gene. Using colony PCR, I could just amplify the DNA from a colony and then show that the transposon increased the size of the expected PCR product compared to a control. Joy. I could also use colony PCR to screen DNA [more...]

TTC VIDEO – Major Transitions in Evolution

New technologies, such as DNA analysis, together with the latest fossil discoveries, make this a fast-changing subject that has grown far more detailed and complete than the presentation you may have learned in school. Among the scientific and popular misconceptions that have been overturned are these: * Neandertals: Recently sequenced DNA from Neandertals suggests that they were not an isolated throwback but contributed some genes to the modern human lineage. Genetic and other clues show that behaviorally Neandertals were very human-like. * Ardipithecus: The 2009 discovery of a relatively [more...]

These are just two recent cases in the US but the implication on DNA sequence patents may be far reaching. Back in 2000, when the new economy was still alive, there was a lot of focus on DNA sequence patents. A lot of companies were very cautious and a lot of genetic test developments were stalled because of patent issues. When asking an expert you could get philosophical answers: either you licence or you ignore or you litigate. Now in 2000 there were still patents from the early days of DNA sequencing around. Some of these patents might have been inventive as the inventive step was a lot [more...]

The court was told that DNA samples taken from a seatbelt, in what is believed to have been the getaway car, and the tip of a latex glove that was found in the vehicle, match DNA samples taken from Duffy and therefore link him to the car. An apparent attempt to burn the car, which had been bought 13 days before the attack through Autotrader for £380, had failed. A forensic scientist told the court that the chance of the DNA found on the latex glove belonging to any other person but Duffy was one in a billion. A robust universal method for extraction of genomic DNA from ...

The intactness of DNA is the keystone of genome-based clinical investigations, where rapid molecular detection of life-threatening bacteria is largely dependent on the isolation of high-quality DNA. Various protocols have been so far developed for genomic DNA isolation from bacteria, most of which have been claimed to be reproducible with relatively good yields of high-quality DNA. Nonetheless, they are not fully applicable to various types of bacteria, their processing cost is relatively high, and some toxic reagents are used. The routine protocols for DNA extraction appear to be [more...]

Question by bob oso: Is there a different type of primer i have to use when running PCR with genomic DNA compared to cDNA? Im still in school but when i run a PCR, I normally isolate RNA first then RT to cDNA and run PCR with that. This time i isolated the genomic DNA from the sample and ran PCR using the beta-actin primer i always used. Dimers were faintly visible on my gel but nothing really came up like it normally would have or should have. I was told that when running PCR with genomic DNA, a different kind of primer had to be used; one that is specific to genomic DNA. Im confused as to [more...]

Paternity Blood Test | Diet and Health Tips & Products Reviews

Today, the most accurate tests (99.99%) to determine the paternity of a child are made through DNA. Collecting DNA sample is done two ways: Buccal swab (swabbing the inside of the cheek) and blood samples. Known as PCR (polymerase chain reaction), the procedure is used in comparing samples of the childís DNA with that of the alleged father. Since the childís DNA is a fairly equal combination of DNA of his parents, excluding the DNA of the mother gives us a genetic profile of the biological father. However, before science knew how to isolate DNA, blood samples were the standard in a variety [more...]
